Modified V.P. Broth is recommended for performing VP test of Bacillus cereus from food samples. Proteose peptone in the medium provides nitrogenous nutrients. Bacillus cereus gives positive reaction for V.P. Test. Acetyl methyl carbinol is produced from glucose by B.cereus. This is determined by adding 0.2 ml of 40% potassium hydroxide (R030) and 0.6 ml of 5% alcoholic 1-naphthol (R029) solution to 1.0 ml of 18-24 hour old culture tube. On addition of few crystals of creatine the reddish pink to purple colour develops within 15 minutes.
